Embedding

This is a process of block making, it is done in embedding box. Counting two ‘L’ shape piece of matel the one arm of ‘L’ is longer then the other two LR joint a for a rectangular box with acts as cast for the tissue block.L is also known as ‘Leuchard mould’.

 Process of block making

Two LR joint on a sheet of mettle or glass surface. A little of molten p[araffin in pourd in the “L” square and is allow to cool down so that “L” doesnot get disturubed during tissue embedding. The tissue from paraffin bath is picked up with a tissue forcep and is place in the centre of “L” square. Molten wax is known poured in the square molten slowly to the filled the square up to surface it is left undisturbed for over one hours until the paraffin cools down and becomes head. Limbs of “L” are removed the paraffin is detached from the glass surface known this block is ready for section cutting.

Before section cutting knif of microtome should be made sharp. This is done a process as honing and stropping.

Honing :- It means grinding of edge knife on a special stone. The hon is a rectangular block of stone. It has two sides the coarse. When is edge is buddy nicked Fine side is used when edge has minner irragularatory the length of hon should be to present whole of the stone. Should sufficient in support the knife.

Honing techanequic :- Clean the knife with xylene socked cloth put on the hon back a damp.

It from sleeping during the process of honing. Clean the surface of hon and lubricate it with coconet oil. Place the knife at one end of the hon with edge facing away from you. Push the knife diagonally forward with the cutting edge leading. While done this keep the knife flat to the hon by its own weight under continue light processure.

The process of forward moved and backward move of knife should be continue for 10 minute for cycle. The edge knife it is appears to be sharp and smooth. Stropping is start it theedage is stillirregular and blunt honing is repeted for the second time for another 10 minutes.

Stropping :- Stropping is a process of removing irregulatory edge of flexable lether strop it helps removing dullness of edge of the knife. The movement of the knife of the stropping should be indirection oppsite that of honing. It should not have dust or absorbing material on its movement during stropping should be rhythmic and study 22-30 dubble stropin sufficient.

Rericulocyte Staining process

Rericulocyte Staining process A reticulocyte stain is a technique a day of Highlight RNA in reticulocytes, A type oc Immuture RBC. This stain commonly use in Hematology department in pathology lab. Edta sample mixed with Reticulocyte staining Solution(Methylene blue) ↓ After Mixed incubate at room temperature. ↓ Prepare   a blood smears ↓ Wait A slide Air-dried the smear ↓ Examined under microscope ↓ The number of reticulocyte count in the patient blood sample is countyed and expressed a percentage   The stain show the RNA in reticulocytes as a blue colour against the pink colour of the rest of the cell. The pattern of the blue colour indicatye the stage of the reticulocyte A hight reticulocyte count can indicate that immutur red blood cells are not surviving longenough, & that the bone marrow needs to increase production. The reticulocyte count is an important role of monitoring of red blood cellssurvival & formation activity o...

MICROSCOPE

MICROSCOPE Introduction of microscope The microscope is indispensable for a clinical laboratory technician. It is very important to have a sound knowledge of the microscope which is going to be a constant comparing in his daily work. To obtain desired magnification for visualizing the objects, the scientists did untiring efforts. The world microscope is derived from two Greek words - ''micro'' (small) and ''scope'' (view). Simple microscope discoveries by Anthony von Leeuwenhoek (1632-1723) and subsequently compound microscope by Robert Hook (1635-1703) were made. It is an instrument by which very small things can be seen which are not seen by the necked eye. This is done by producing an enlarged and well defined image of the objects by a system of lens. Principle of microscope The light beam, which falls on the object, p...
